In the sense of female dog as well as the metaphorical sense. Is "κύων" used for both masculine and feminine?

Yes.  The word is made feminine through the use of the article (ἡ κύων) or an adjective with a feminine ending (e.g. κύων τις πονηρά).  But I don't believe it ever had our metaphorical sense in ancient Greek.  Which reminds me, Congrats on the book!  (-;
